گزینش ژنوتیپ‌های برتر باقلا (Vicia faba L.) از لحاظ عملکرد و مقاومت به بیماری با استفاده از روش GYT بای‌پلات

چکیده

سابقه و هدف: افزایش سطح مقاومت در برابر بیماری‌های لکه برگی باقلا، لکه شکلاتی (Botritis fabae)، لکه برگی آلترناریا (Alternaria alternate)، سوختگی استمفیلیومی S. botryosum) و (Stemphylium vesicarium، از اهداف اصلی برنامه‌های اصلاحی باقلا هستند. مقاومت به بیماری، زمانی بیشترین کارایی را دارد که با عملکرد دانه بالا در یک ژنوتیپ همراه باشند. روش تجزیه ژنوتیپ بر اساس بای‌پلات ژنوتیپ در عملکرد×صفت (GYT) به انتخاب ژنوتیپ‌هایی مطلوب برای صفات مورد بررسی در حضور عملکرد دانه بالا کمک می‌کند.
مواد و روش‌ها: آزمایش در قالب طرح بلوک‌های کامل تصادفی با سه تکرار در دو سال زراعی ۱۳۹۷ تا ۱۳۹۹ در ایستگاه تحقیقات کشاورزی گرگان اجرا شد. پانزده ژنوتیپ باقلا شامل چهار رقم تجاری (G11 تا G14)، یک شاهد حساس به بیماری (G15) و ده لاین امیدبخش (G1 تا G10) از برنامه ملی اصلاح باقلا مورد ارزیابی قرار گرفتند. صفات مورد مطالعه شامل شدت بیماری، سطح استانداردشده زیر منحنی پیشرفت بیماری (sAUDPC)، ارتفاع گیاه، اجزای عملکرد و عملکرد دانه بود. جهت محاسبه sAUDPC، پیشرفت بیماری در پنج مرحله از زمان ظهور علائم نمونه‌برداری شد. داده‌ها با نرم‌افزار SAS تحلیل و میانگین‌ها با آزمون حداقل اختلاف معنی‌دار (LSD) در سطح احتمال یک درصد (P≤0.01) مقایسه شدند. برای ارزیابی همزمان مقاومت به بیماری و بهبود صفات کمّی، تحلیل دوبعدی GYT در ترکیب با تجزیه خوشه‌ای به‌کار گرفته شد تا بهترین ژنوتیپ‌ها انتخاب و نقاط قوت و ضعف هر ژنوتیپ از طریق ترکیب عملکرد با سایر صفات هدف تعیین شود.
یافته‌ها: نتایج تجزیه واریانس مرکب نشان داد که تفاوت بسیار معنی‌دار در سطح احتمال یک درصد بین ژنوتیپ‌های باقلا برای صفات زراعی و مقاومت به بیماری وجود دارد. میانگین عملکرد دانه در بین ژنوتیپ‌ها ۳۳۲۴ کیلوگرم در هکتار و دامنه تغییرات عملکرد بین 7/4461-1786 کیلوگرم در هکتار متغیر بود. یافته‌ها نشان داد، بای‌پلات ژنوتیپ در عملکرد × صفت، 6/88 درصد از کل تغییرات را توجیه‌ می‌کند. نمای کدام ژنوتیپ در کجا در بای‌پلات چندضلعی، فضای بای‌پلات را به شش بخش تقسیم کرد که سه بخش آن حاوی ترکیبات مطلوب عملکرد-صفت بودند. در بخش اول، ژنوتیپ G12 (رقم فیض) همراه با ترکیب عملکرد با صفات تعداد دانه در غلاف و وزن صد دانه قرار گرفت. در بخش دوم، ژنوتیپ G10 (لاین HBP/SOE/99) در ترکیب با صفات ارتفاع بوته و تعداد شاخه متمایز شد. در بخش سوم نیز ژنوتیپ‌های G13 (رقم شادان) و G14 (رقم مهتا) به دلیل داشتن شاخص‌های برتر مقاومت به بیماری در رأس چندضلعی قرار گرفتند. بر اساس نمایش برداری GYT بای‌پلات، مقادیر sAUDPC بیماری سوختگی استمفیلیومی و مقادیر sAUDPC بیماری لکه برگی آلترناریایی در ترکیب با عملکرد دانه بیشترین همبستگی را داشتند. نمای مختصات میانگین تستر (ATC) از GYT بای‌پلات، 15 ژنوتیپ را به عنوان ژنوتیپ‌های با عملکرد بهتر از میانگین طبقه‌بندی کرد و به صورت G13 > G10 > G14 > G12 > … > G15 رتبه‌بندی شدند، در حالی که G10 و G12 با تمام ترکیبات مثبت عملکرد-صفت به عنوان بهترین ژنوتیپ‌ها و ژنوتیپ‌های G5، G7 و G15 به عنوان ضعیف‌ترین ژنوتیپ‌ها رتبه‌بندی شدند. بر اساس تجزیه خوشه‌ای، ژنوتیپ‌های باقلا به چهار گروه تقسیم شدند. گروه‌های متمایز به‌دست‌آمده، برای استخراج ژنوتیپ‌هایی با ویژگی‌های متنوع و شناسایی تنوع خزانه ژنی مفید هستند.
نتیجه‌گیری: در برنامه‌های اصلاح‌نژادی باقلا، تلفیق صفاتی چون طول غلاف، وزن صد دانه و مقاومت به بیماری با عملکرد بالای دانه در یک ژنوتیپ واحد از اهمیت ویژه‌ای برخوردار است. این مطالعه نشان داد که GYT بای‌پلات ابزاری کارآمد برای شناسایی ژنوتیپ‌های ایده‌آل است که تعادل مناسبی بین عملکرد و چندین صفت زراعی برقرار می‌کنند. در نهایت ژنوتیپ‌‌ G10 (HBP/SOE/99) ترکیب مطلوبی از مقاومت به بیماری‌های قارچی، پتانسیل عملکرد بالا و سازگاری با شرایط اقلیمی منطقه گرگان داشت و به‌عنوان رقم جدید یا به‌عنوان والد در برنامه‌های اصلاحی پیشنهاد می‌گردد.

Background and objectives: Enhancing resistance to foliar diseases, particularly chocolate spot (Botrytis fabae), Alternaria leaf spot (Alternaria alternata), and Stemphylium blight (Stemphylium botryosum and S. vesicarium), is a primary objective of faba bean breeding programs. Disease resistance is most effective when integrated with high grain yield within a single genotype. The genotype-by-yield×trait (GYT) biplot method facilitates the identification of superior genotypes that combine high yield with favorable secondary traits.
Materials and Methods: Field experiments were conducted during the 2018–2020 growing seasons at the Gorgan Agricultural Research Station, Iran, using a randomized complete block design (RCBD) with three replications. Fifteen faba bean genotypes, including four commercial cultivars, one disease-susceptible check, and ten promising breeding lines from Irans national breeding program, were evaluated. Traits assessed included disease severity, standardized area under the disease progress curve (sAUDPC), plant height, yield components, and grain yield. Disease progression was monitored at five time points to calculate sAUDPC. Data were analyzed using SAS software, and means were compared using the LSD test at P ≤ 0.01. GYT biplot analysis, integrated with cluster analysis, was employed to simultaneously evaluate disease resistance and yield-related traits for optimal genotype selection.
Results: Combined analysis of variance revealed highly significant differences (P ≤ 0.01) among genotypes for all agronomic and disease resistance traits. The mean grain yield was 3324 kg ha⁻¹, with a range of 1786 to 4461 kg ha⁻¹. The GYT biplot explained 88.6% of the total variation. The "which-won-where" polygon view partitioned the biplot into six sectors, three of which highlighted favorable trait combinations. Sector 1 contained genotype G12 (Feyz cultivar), combining yield with the number of seeds per pod and 100-seed weight. Sector 2 featured genotype G10 (line HBP/SOE/99), combining yield with plant height and number of branches. Sector 3 was dominated by genotypes G13 (Shadan) and G14 (Mahta), which exhibited superior disease resistance indices at the polygon vertices. The vector view indicated strong associations between grain yield and sAUDPC values for Stemphylium blight and Alternaria leaf spot, highlighting the complex trade-offs between yield and disease susceptibility. The average tester coordinate (ATC) view ranked the genotypes as G13 > G10 > G14 > G12 > … > G15. Genotypes G10 and G12 were identified as the best performers due to their positive yield-trait combinations, whereas G5, G7, and G15 were the weakest. Cluster analysis grouped the genotypes into four distinct clusters, providing a basis for diversifying heterotic pools in future breeding efforts.
Conclusion: In faba bean breeding, integrating disease resistance and key yield components (such as pod length and 100-seed weight) with high grain yield in a single genotype is crucial. The GYT biplot proved to be an effective tool for identifying ideal genotypes that balance yield and multiple agronomic traits. Overall, genotype G10 (HBP/SOE/99) demonstrated an outstanding combination of fungal disease resistance, high yield potential, and excellent adaptation to the local conditions of Gorgan, making it a promising candidate for cultivar release or as a parental line in breeding programs.
