However, its protein content is relatively low compared to other cereals. Seed storage protein in rice classified into four groups: albumin and globulin in aleurone layer, and prolamine and glutelin in endosperm. Therefore, during the bleaching process of rice grains, albumin proteins and part of globulin are removed by its bran. Since the protein content of the grain and its amount will be directly related to its nutritional quality and due to the importance of rice grain in diet, seed storage proteins quality in a quantitative and qualitative breeding population of rice such as two new mutant cultivars, Roshan and Shahriar, with their parents (Nemat and Amol3 respectively) as well as qualitative varieties, Sang-tarom, along with two different varieties of brown and black rice was studied.Materials and methods: Qualitative properties of grain proteins were evaluated based on their solubility as well as the seed storage proteins For albumin, globulin, prolamine and glutelin proteins. Quantification of stored proteins was performed by Bradford reagent and quantification of their subunits by densitometric analysis.Results: The results showed that there is a significant difference for the amount of seed storage proteins of different rice genotypes. So that Sang-tarom and Roshan cultivars had the highest amount of albumin protein (19.53 and 22.40 mg/g respectively). This was while the brown grain variety with the lowest amount of albumin and globulin protein and the highest amount of glutelin and prolamine proteins has the highest amount of total protein (1640.43 mg/g). Evaluation of the banding pattern of stored storage proteins also showed that this banding pattern was the same in different genotypes except in Roshan (60 kDa) and Shahriar (13 kDa) mutant cultivars. Densitometric analysis of protein subunits also showed that the highest amount of glutelin subunits was present in Sang-tarom cultivar and the lowest in Shahriar cultivar and brown grain variety. Also, the highest amount of prolamine subunits was observed in Sang-tarom cultivar and brown grain variety and the lowest in Shahriar cultivar. As Shahriar cultivar was able to have the highest amount of glutelin to prolamine protein.Conclusion: Although the profiles of seed storage proteins in different varieties of rice are the same, but this pattern is different in the two cultivars of mutant Roshan and Shahriyar. So that, the absent of the 60kDa subunit of proglutelin and the 13kDa subunit of prolamine can be used as protein markers in both Roshan and Shahriar, respectively. Our study demonstrated that although there is a significant relationship between protein subunits and the amount of stored proteins, but there was no significant relationship between stored proteins with different quantitative and qualitative rice cultivars. Therefore it seems that improving the seed storage protein quality will probably not limit the improvement of new rice cultivars with desirable quantitative and qualitative characteristics.}, keywords = {Rice,Storage proteins,Band patterns of seed storage protein}, title_fa = {ارزیابی کیفیت پروتئین‌های ذخیره‌ای دانه در برخی از ارقام کمی و کیفی برنج (Oryza sativa L.)}, abstract_fa = {سابقه و هدف: برنج منبع مناسبی از انرژی محسوب می‌شود و از بین پروتئین‌های گیاهی بالاترین کارایی را برای ارزش غذایی پروتئین به خود اختصاص داده است. این درحالی است که محتوای پروتئین آن در مقایسه با سایر غلات نسبتا پایین می‌باشد. پروتئین‌های ذخیره‌ای دانه برنج شامل آلبومین و گلبولین در لایه آلئورون و پروتئین‌های پرولامین و گلوتلین در آندوسپرم دانه می‌باشند. بنابراین طی فرایند سفید کردن دانه برنج پروتئین‌های آلبومین و بخشی از گلوبولین در جداسازی سبوس آن حذف می‌گردند. باتوجه به اهمیت مصرف دانه برنج در رژیم غذایی، در این تحقیق کیفیت پروتئین‌های ذخیره‌ای دانه در یک جمعیت اصلاحی کمی و کیفی برنج شامل دو رقم جدید موتانت روشن و شهریار و والد آن‌ها (به ترتیب ارقام نعمت و آمل3) به همراه رقم کیفی سنگ طارم و دو واریته مختلف برنج با رنگ دانه قهوه‌ای و سیاه مورد مطالعه قرار گرفت.مواد و روش‌ها: ارزیابی خصوصیات کیفی پروتئین‌های دانه بر اساس قابلیت انحلال آن‌ها (برای پروتئین‌های آلبومین، گلوبولین، پرولامین و گلوتلین) و همچنین بر اساس الگوی باندی پروتئین‌های دانه انجام شد. کمیت سنجی پروتئین‌های ذخیره‌ای با استفاده از معرف برادفورد و کمیت سنجی زیرواحدهای آن‌ها نیز با آنالیز دنسیتومتری صورت گرفت.یافته‌ها: نتایج این مطالعه نشان داد که اختلاف معنی‌داری برای میزان پروتئین‌های ذخیره‌ای در ژنوتیپ‌های مختلف دانه برنج وجود دارد. بطوریکه ارقام سنگ طارم و روشن بیشترین میزان پروتئین آلبومین را به خود اختصاص داده بودند (به ترتیب 53/19 و 40/22 میلی‌گرم بر گرم). این درحالی بود که واریته دانه قهوه‌ای با کمترین میزان پروتئین آلبومین و گلوبولین و بیشترین میزان پروتئین‌های گلوتلین و پرولامین بیشترین میزان پروتئین کل را به خود اختصاص داده بود (43/1640 میلی‌گرم بر گرم).ارزیابی الگوی باندی پروتئین‌های ذخیره‌ای نیز نشان داد که این الگوی باندی به جز در ارقام موتانت روشن (kDa 60) و شهریار (kDa 13) در دیگر در ژنوتیپ‌های مختلف مورد ارزیابی یکسان و مشابه بود. آنالیز دنسیتومتری زیرواحدهای پروتئینی نیز نشان داد که بیشترین میزان زیرواحدهای گلوتلین در رقم سنگ طارم و کمترین آن در رقم شهریار و واریته دانه قهوه‌ای وجود دارد. همچنین بیشترین میزان زیرواحدهای پرولامین نیز در رقم سنگ طارم و واریته دانه قهوه‌ای و کمترین آن در رقم شهریار مشاهده شد. بطوریکه رقم شهریار توانسته بود بیشترین میزان پروتئین گلوتلین به پرولامین را به خود اختصاص دهد. نتیجه‌گیری: نتایج این تحقیق نشان داد که اگرچه پروفایل پروتئین‌های دانه در واریته‌های مختلف کمی و کیفی برنج یکسان و مشابه است، اما این الگو در دو رقم موتانت روشن و شهریار متفاوت می‌باشد، بطوریکه عدم بیان زیرواحدهای kDa 60 پروگلوتلین و زیرواحد kDa 13 پرولامین می‌توانند به عنوان مارکرهای پروتئینی به ترتیب در دو رقم روشن و شهریار مورد استفاده قرار گیرند. همچنین این نتایج نشان داد که اگرچه بین زیرواحدهای پروتئنی و میزان پروتئین‌های ذخیره‌ای ارتباط معنی‌داری وجود دارد، اما بین پروتئین‌های ذخیره‌ای با ارقام مختلف کمی و کیفی برنج ارتباط معنی‌داری حاصل نشد، بنابراین به نظر می‌رسد که احتمالا اصلاح کیفیت پروتئین‌های دانه محدودیتی را برای اصلاح ارقام جدید برنج با خصوصیات کمی و کیفی مطلوب ایجاد نخواهد کرد.}, keywords_fa = {برنج,پروتئین‌های ذخیره‌ای,الگوی باندی پروتئین‌های دانه}, url = {https://ejcp.gau.ac.ir/article_6266.html}, eprint = {https://ejcp.gau.ac.ir/article_6266_0669206d101221d9046eeed70488b1e6.pdf} }
